In terrible conditions at Sherburn, Staithes were a goal to the good midway through the first half when one of their forwards produced a fine half-volley.
Pier equalised 25 minutes into the second half, Martin Cooper's inswinging corner being help
ed into the net by the Staithes keeper.
Pier boss Paul Romans was preparing for extra-time but in the dying seconds of the game the ball broke to Will Jenkinson and he did the rest.
Romans said: "It was really difficult to play in those conditions so credit to the lads for their efforts and the fact that they kept going right to the end.
"I was all set for extra-time so was delighted when Will scored."
Edgehill 3 (Gill, Jenkinson, White) Snainton 0
Challenge Cup holders Edgehill started their defence of the trophy in style with a comfortable home victory.
Barry Gill fired Edgehill ahead after just 10 minutes and that proved the only goal of the first half.
The hosts increased their lead early in the second half through Luke Jenkinson, and Danny White wrapped things up for Alec Coulson's side with a header from a Lee Lee Cappleman corner.
Reeth 1 Seamer Sports 6 (Jewitt 4, Ogle 2)
New signing Rich Ogle scored two in the first five minutes of his Seamer debut.
Ogle's early brace put Seamer well in control and they were three goals up at half-time with Dan Jewitt scoring his first of the afternoon.
The same player secored another three in the second half and although the hosts pulled one back Seamer were always on top.
Eastfield 8 (Hall 5, C Dove, Dobson, Warnett)

Striker Eric Hall went nap as he bagged five quality goals to help Eastfield progress into the second round of the competition.
He was joined on the scoresheet by Cameron Dobson, Chris Dove and Phil Warnett in a largely one sided affair.
Newlands Park 2 (G Thomas, Laing) Lealholm 3
Lealholm were two goals to the good at the interval and added a third early in the second half.
Gary Thomas pulled a goal back for Geoff Nock's outfit with a fine lobbed effort and Ian Laing was also on target for the hosts.
Westover 2 (Joe Cairns, Jon Cairns) Bedale 5
Brothers Joe and Jon Cairns were once again on target for Westover but they found it hard work against a strong Bedale side.
Bedale were 3-0 up when Joe found the target with a neat finish.
